血清12项细胞因子在冠心病诊断中的应用价值 被引量:12

Studies on twelve cytokines in serum of coronary heart disease patients

摘要 目的观察冠心病患者血清多种细胞因子的变化及相互关系,以探索血清细胞因子谱在冠心病诊断中的应用价值。方法观察76例确诊的冠心病患者及26名健康查体者血清细胞因子水平,利用 RANDOX 细胞因子芯片和 EVIDENCE 全自动芯片分析仪测定血清12项细胞因子,运用生物信息学方法绘制血清细胞因子图谱。结果冠心病患者血清白细胞介素(IL)-1β、IL6、IL8、肿瘤坏死因子(TNF)-α水平分别为(4.32±8.14)ng/L,(13.16±16.81)ng/L,(375.53±292.14)ng/L,(15.46±15.38)ng/L,其浓度显著高于正常人群(P<0.01)。IL-4水平为(0.65±1.51)ng/L,低于正常人群(P<0.05)。稳定型心绞痛、不稳定型心绞痛和急性心肌梗死组患者血清中 IL-1β、IL-4、IL-6、IL-8、血管内皮生长因子(VEGF)、TNF-α、单核细胞趋化蛋白-1(MCP-1)、表皮生长因子(EGF)等均呈不同程度变化,特别是 IL-6、IL-8、VEGF、TNF-α水平变化幅度最大。结论血清多种细胞因子在冠心病不同分型中呈规律性变化,提示细胞因子谱在冠心病的诊断及预后具有潜在的应用价值。 Objective The significance of serum cytokine profile in coronary heart disease patients have been evaluated by observing the variation cytokines Methods The serum IL-1α,IL-1β, IL-2, IL-4, IL-6, IL-8, IL-10, VEGF, IFN-% TNF-α, MCP-1 and EGF in 76 patients and 26 matched healthy volunteers have been observed. Serum cytokines had been measured by protein chips methods in EVIDENCE 180 automatic biochips analyzer and cytokine profile were got by bioinformatics. Results The serum level of IL-1β,IL-6, IL-8,and TNF-α were (4. 32 ± 8. 14) ng/L , (13. 16 ±16. 81 ) ng/L, (375. 53 ± 292. 14) ng/L, ( 15.46± 15.38) ng/L respectively, which increased obviously in patients ( P 〈 0. 01 ). The serum level of IL-4 decreased compared with the control group and the level was (0. 65 ± 1.51 ) ng/L ( P 〈 0.05 ) .There were many abnormal serum cytokines have been found in three different types of patients especially the serum level of IL-6, IL-8, VEGF and TNF-α. Conclusion The variation of serum cytokines is related with the different types of coronary heart disease, which suggests that cytc.kine profile is useful to diagnose and follow up these patients.
