冷暴露大鼠心肌线粒体内膜流动性及寡霉素敏感蛋白变化

Changes of Intimal Fluidity and Function of the Oligomycin Sensitive Protein of Myocardial Mitochondria in Rats During Cold Exposure

摘要 作者以测定无机磷的释放比较线粒体H运转ATP酶对寡霉素的敏感性,藉1,6-二苯基、1,3,5-已二烯探针比较线粒体内膜的流动性。发现:(1)线粒体H运转ATP酶的寡霉素抑制率在正常对照组为99.00±0.20%,冷暴露1,2,3,4和5周分别为:66.71±0.45%,59.51±0.53%,52.01±0.83%,69.32±0.91%和80.63±0.63%。(2)线粒体内膜荧光偏振度在对照组为0.292±0.001,冷暴露1,2,3,4和5周分别为:0.283±0.002,0.278±0.001,0.263±0.001,0.268±0.001和0.272±0.001。结果表明:冷暴露后大鼠心肌线粒体H^+运转ATP酶对寡霉素的敏感性降低。线粒体内膜的流动性增强。 The sensitivity of mitochondrial H -transporting ATPasc to oligomycin was assayed by releasing of phosphorus. The membrane fluidity of mitochondria was compared by using fluorescent probe 1,6- dipheny 1-1,3,5-hexatricne. It was found that the rate of inhibition by oligomycin of mitochondrial H - transporting ATPase from the rats of the control was 99.00±0.2%; the rates of inhibition by oligomycin of mitochondrial H -transporting ATPase from the rats exposed to the cold for 1, 2, 3, 4 and 5 weeks were 66.71 ±0.45%, 59.51±0.53%, 52.01±0.83%, 69.32 ±0.91% and 80.63±0.63% respectively. The fluorescence polarization of submltochondria from the rats of the control was 0.292 0.001; and those of submitochondria from the rats exposed to the cold for 1, 2. 3, 4 and 5 weeks were 0.283 ±0.002, 0.278±0.001, 0.263±0.001, 0.268± 0.001, 0.272±0.001 respectively. It was shown that the sensitivity to oligomydn of submitochondnal H - transporting ATPasc was decreased after cold exposure . The fluidity of submitochondnal membrane in the rats during cold exposure is obviously higher than that of the control.
