应用PCR-SSCP法检测乳腺癌患者血中p53基因突变 被引量:5

摘要 [目的]探讨乳腺癌患者外周血p53基因突变的临床意义.[方法]应用聚合酶链反应-单链构象多态性分析(PCR-SSCP)的方法检测35例乳腺癌患者和20例良性肿瘤患者外周血中p53基因外显子5~8的突变情况.[结果]乳腺癌患者p53基因突变的检出率为40.0%(14/35),显著高于对照组良性肿瘤患者的5.0%(1/20)(P<0.05).[结论]p53基因突变在乳腺癌外周血中有较高的检出率,是乳腺癌发生的早期信号. [Objective] To investigate the clinical significance about p53 gene mutation in blood of the patients with breast cancer. [Method] Use of single strand conformation polymorphism analysis of polymerase chain reaction products(PCR - SSCP) to inspect p53 gene mutation from 35 blood samples of the patients with breast cancer and 20 blood samples of the patients with breast benign tumor. [Result] The mutation rate of the patients with breast cancer were 40.0% (14/35), and the mutation rate of the patients with breast benign tumor were only 5.0% (1/20). The difference was significant (P 〈 0.05). [Conclusion] Blood p53 gene mutation from the patients with breast cancer have higher detection rate, which is the early period signal of breast cancer.
