雌孕激素对人卵巢颗粒细胞瘦素受体表达的影响 被引量:1

摘要 目的通过人卵巢黄素化颗粒细胞体外培养,观察雌、孕激素对其瘦素受体蛋白表达的调控作用。方法行体外受精-胚胎移植患者经阴道超声引导下取卵时留取卵泡液,分离颗粒细胞体外培养,培养至第3天加入不同浓度的17-β雌二醇或孕酮,继续培养2d,采用免疫细胞化学方法将颗粒细胞进行荧光标记,经流式细胞式检测其瘦素受体蛋白表达阳性细胞百分率。结果流式细胞仪检测发现所检测各浓度17-β雌二醇、孕酮均能增强颗粒细胞瘦素受体蛋白的表达,17-β雌二醇作用高峰浓度为1μg/ml,孕酮作用高峰浓度为1ng/ml。结论雌孕激素在体外可增强人卵巢黄素化颗粒细胞瘦素受体蛋白的表达,提示瘦素可能参与卵巢功能的调节,为深入研究瘦素在卵巢中的作用提供理论依据。
