探讨硫酸软骨素酶在兔耳增生性瘢痕模型中的作用 被引量:4

摘要 目的应用兔耳模型探讨硫酸软骨素裂解酶在增生性瘢痕中的作用。方法将健康的日本大耳白兔12只,随机分成胶原酶治疗组(A组)、硫酸软骨素裂解酶与胶原酶混合治疗组(B组)、硫酸软骨素裂解酶治疗组(C组)、对照组(D组)。采用复合染色法观察组织形态学变化,并进行图像定量分析,羟脯氨酸含量测定。结果各治疗组酸性黏多糖染色变浅,数量减少,胶原纤维被破坏,排列稀疏。A、B、C、D组的羟脯氨酸含量,随着时间的变化其含量不一致(P<0.05),A、C组与B组比较在第28天差异有显著意义(P<0.05),B组与A组比较在第35天差异有显著意义(P<0.05),在用药两周后B组羟脯氨酸含量始终低于其他各组。结论硫酸软骨素裂解酶水解了硫酸软骨素,在去除了胶原纤维周围的酸性黏多糖后,促进了胶原的降解,提示可能为瘢痕的治疗提供新的途径。 Objective To investigate the effects of chondroitinase on the formation of hypertrophic sear in rabbit ears. Methods Wound - healing models were established on ears of 12 healthy the Jepanes rabbits, which were randomly divided into the collagenase- treated group (group A), the chondroitinase ABC and collagenase - treated group (group B), the chondroitinase ABC- treated group (group C) and saline - treated group (group D ). Compound staining was used to observe the morphological and histological changes in hypertrophic scar. Image analysis technique and chemical way were employed to measure the contents of collagen and hydroxyproline. Results It was found that the colour of the acid mucopolysac- charide in each control group was turned into pale blue, the collagen fibers in hypertrophic scar arranged sparsely and destroyed greatly according to compound staining. Hydroxyproline content of the A, B, C and D were significandy different as time went by (P 〈 0.05). Compared with group A and C, hydroxyproline content of group B had significant difference on the 28th day (P 〈 0.05), and there was significant difference on the 35th day between group B and group A (P〈0.05), the hydroxyproline content of group B was much lower than those of other groups after two weeks' treating (P〈0.05). Conclusion Chondroitinase A, B, C increase the degradation of collagen fibers after removing the acid mucopolysaccharide around the collagen fibers. This may provide a new way on hypertrophic scar treatment.
