不同化疗药物对宫颈癌裸鼠移植瘤微小染色体维持蛋白7表达影响的研究 被引量:1

摘要 目的:研究铂类和氧化嘧啶类药物对HeLa细胞裸鼠移植瘤微小染色体维持蛋白7表达的影响。方法:将HeLa细胞注射于4~6周龄雌性裸鼠皮下建立移植瘤模型,根据所用药物将裸鼠分组为A:对照组、B1:顺铂组、B2:卡铂组、B3:5-FU组、B4:卡培他滨组(capecitabine,商品名:xeloda)、B5:顺铂+5-Fu组、B6:顺铂+卡培他滨组、B7:卡铂+5-Fu组。RT—PCR法检测微小染色体维持蛋白7(MCM7)的表达,并与大体组织的观察指标如肿瘤体积、质量的变化及抑瘤率进行比较。结果:B1、B2、B5、B6、B7的MCM7表达低于A,差异有统计学意义(P〈0.05),B7的MCM7表达低于B4,差异也具有统计学意义(P〈0.05),但析因分析未显示出他们有正交互效应。结论:对宫颈癌移植瘤抑制作用强的药物其MCM7的表达低,MCM7有可能成为评价宫颈癌化疗药物疗效的生物学指标。 Objective:To study the the inhibition somal maintenance 7 ( MCM7 ) of cervical cancer xenografts of expression level of minichromo- treated with platinum and fluoroprimidine drugs. Methods:The HeLa cells were subcutaneous injected into the female nude mice of 4 - 6 weeks of age. The mice were divided into 8 groups according to the drugs, namely, A : control group, B1 : cisplatin, B2 : carboplatin, B3 : 5-FU, B4 : capecitabine, B5 : cisplatin + 5-FU, B6:cisplatin + capecitabine, B7: carboplatin + 5-FU. MCM7 expression was evaluated by RT- PCR, and was compared to the parameters, such as the volume and weight of the tumors and the inhibition ration. Results: The MCM7 expression of group B 1. B2. B5. B6. B7 was significantly less than that of control group( P 〈 0.05 ). On the other hand ,the MCM7 expression of group B7 was also significantly less than that of B4 ( P 〈 0.05 ). However, there was no interaction effect between B7 and B4 by muhifactofial analysis. Conclusion :The drug that had stronger inhibition effect might be had lower expression of MCM7, MCM7 may be a new indicator to evaluate the curative effect of chemotherapy on cervical cancer.
