转野生荠菜凝集素基因棉花对棉蚜的抗性鉴定 被引量:5

摘要 利用人工网室对45个转野生荠菜凝集素(WSA)基因棉花工程植株进行抗棉蚜鉴定,结果表明,38个转WSA基因棉花工程植株对棉蚜虫口密度的控制效果达到65%以上,蚜害级别均为Ⅰ级。南繁种植转化一代,标记基因(卡那霉素抗性基因)检测结果,38个株行抗、感植株分离比例符合3:1,WSA基因是以单一位点整合到棉花染色体组中,不存在多个位点的整合方式;纤维品质测定结果,其中的9个株行达到高品质棉标准。与受体品种相比,9个高品质转WSA基因棉花株系对棉蚜虫口密度的控制效果达65.23%-74.39%,蚜害指数减退率皆达到80%以上,对棉蚜的抗性达到高抗水平。并对转WSA基因棉花植株上未出现死亡棉蚜的原因进行了讨论。 Resistance of 45 trans- wild shepherd's purse agglutinin (WSA) gene cotton engineering plants to aphis was identified by aphis natural immigration in artificial net - room. The results indicated that the repression effects of 138 trans - WSA gene plants on aphis density were above 65%, the aphis damaging grades of 38 plants were all grade Ⅰ. First generation plants of genetic transformation(T1 ) were planted in Hainan island from winter of 2003 to spring of 2004, NPTⅡ gene as marker gene, coding Kanamycin resistance, was screened in 38 T1 populations, the results indicated that the segregation proportion of resistant plants to susceptible plants in T1 population accords with 3: 1, the WSA gene is integrated into cotton genome at single locus, the integration of many loci into cotton genome rarely emerges. Fiber quality test showed that 9 trans - WSA gene cotton plant rows with high quality were obtained in 38 aphis - resistant plants. Furthermore, 9 trans - WSA gene cotton lines and their receptor varieties were identified for resistance to aphis. The results indicated that the repression effects on aphis density of 9 trans -WSA gene lines were 65. 23% -74.39%, and the decreased rates of aphis damaging index of 9 lines surpass 80%, and their resistance to aphis achieved high level, compared with their corresponding receptor varieties. The reason about absence of dead aphis on trans - WSA gene cotton plants is also discussed in this paper.
