实时荧光定量PCR检测耐药结核分枝杆菌中耐药相关基因whiB7的表达 被引量:2

Expression of the whiB7 gene induced by antibiotics in multidrug-resistant mycobacterium tuberculosis and measured by quantitive real time PCR

摘要 目的探讨结核分枝杆菌耐药性与whiB7基因表达水平之间的关系。方法以耐受利福平、异烟肼、链霉素、乙胺丁醇和氟喹诺酮的结核分枝杆菌临床分离株为研究对象,利用实时定量PCR的方法,检测药物刺激结核分枝杆菌后whiB7基因表达水平的变化。结果除利福平刺激相应的菌株后,whiB7基因的表达水平变化不明显外,其余药物的刺激均会引起结核分枝杆菌耐药株的whiB7基因表达水平发生显著变化。结论结核分枝杆菌的whiB7基因是对多种抗生素刺激发生反应的基因,推测whiB7的表达参与结核分枝杆菌的耐药过程。 Objective To investigate the relationship between the expression of whiB7 gene and the multidrug resistance of the mycobacterium tubereulosis(MTB). Methods The antibioticses, including rifampicin, isoniazid, streptomycin, ethambutol and fluoroquinolones, were used to stimulate the clinic MTB accordingly. After the drug stimulation, the expression of whiB7 was measured by quantitive real time PCR. Results The expressing level of whiB7 gene was significantly improved after the MTB being stimulated by isoniazid, streptomycin, ethambutol and fluoroquinolones except for the rifampicin. Conclusions WhiB7 was widely involved in the resistance of the MTB to the many antibioticses.
