环氧化酶-2mRNA在乳腺癌组织中的表达及临床意义

摘要 目的探讨乳腺癌组织中COX-2mRNA的表达与乳腺癌发生、发展及预后的关系.方法以β-action基因为内参照,应用逆转录聚合酶链反应(RT-PCR)技术,检测35例乳腺癌组织、相应癌旁组织、正常乳腺组织中COX-2mRNA的表达,应用Gene Tools from SynGene软件,对RT-PCR产物电泳带进行半定量分析.结果35例癌组织中,29例COX-2表达明显增高(0.83),相应6例癌旁组织COX-2有不同程度的表达(0.17),正常乳腺组织表达很弱或无表达,后二者与癌组织中COX-2表达差异均有显著意义(P<0.05).COX-2表达同年龄、原发肿瘤大小、TNM分期及术前化疗与否无关(P>0.05),而与淋巴结转移状况有关(P<0.05).结论乳腺癌组织中COX-2的表达明显高于相应癌旁组织、正常乳腺组织,且其表达与淋巴结转移状况有关. Objective To discuss the relation between the expression of COX - 2 in breast cancer and the development and prognosis of breast cancer. Methods With? - action gene as reference, the COX - 2 mRNA was monitored in 35 specimens of breast cancer tissues, paried adjacent breast tissues, normal breast tissues by reverse transcription -polymerase chain reaction (RT- PCR) . Results The rate of position COX -2 expression was 0.83 for breast cancer tissues significantly than that for the paired adjacent breast tissues (0. 17) and normal breast tissues ( nearly no expression) ( P 〈 0. 05 ), the expression of COX - 2 was not correlated with the clinical parameters, such as patient' s age, size of primary tumor, P - TNM stage, and therapy before operation ( P 〉0. 05), but correlated with lymph node metastasis ( P 〈 0. 05) . Conclusion COX - 2 overexpression may play a crucial role in the carcinogenesis, development of breast cancer.
