摘要
目的比较利巴韦林等五种药物对汉坦病毒(HV)的体外抑制作用。方法利用HV76-118株感染传代培养的Vero-E6细胞,采用免疫荧光法分别评价五种药物的抗病毒效果。结果五种药物均对汉坦病毒的入侵无阻断作用;利巴韦林对汉坦病毒增殖有明显的抑制作用,其最大无毒浓度(TD0)为500μg/ml、最小有效浓度(MTC)为31.2μg/ml,干扰素α-1b(赛若金)TD0为10万U/ml、MTC为0.5万U/ml,阿昔洛韦TD0为625μg/ml、MTC为156μg/ml,更昔洛韦、膦甲酸钠各浓度均未见对病毒增殖有抑制作用。结论在一定浓度范围内利巴韦林、赛若金、阿昔洛韦对出血热病毒均具有抑制作用。
Objective To compare the antiviral activity of five drugs in vitro. Methods The cultured Veto-E6 cells were infected with HV 76-118. The antiviral activity of five drugs was evaluated by using immunofluorescence assay in vitro. Results These five drugs all had no effect to prevent the invasion of Hantavirus. By immunofluorescence assay, it was found ribavirin had significantly inhibitory effect on the proliferation of Hantavirus with the maximal tolerance concentration (TD0) being 500 μg/ml and the minimal therapeutic concentration (MTC) being 31.2 μg/ml respectively; The TD0 of Sinogen was 10^5 U/ml and the MTC 0. 5 × 10^5U/ml; The TD0 of Acyclovir was 625 μg/ml and the MTC was 156 μg/ml. However, no inhibitory effect was observed in the other concentrations of Foscavir and Ganciclovir. Conclusion In certain range of concentration, Ribavirin, Sinogen and Acyclovir have inhibitory effect on Hantavirus.
