3个miniSTR基因座在高度降解检材中的应用及其遗传多态性 被引量:4

摘要 目的研究D1S1677、D4S2364、D10S1248 3个m in iSTR基因座在DNA高度降解检材中的法医学应用价值并调查河北地区汉族人群3个基因座的遗传多态性。方法对所有样本的3个基因座进行PCR扩增;PCR产物在AB I3100 Avant基因分析仪上电泳分离;GeneScan 3.7及Genotyper3.7软件分析结果。结果3个m in iSTR基因座均获得了清晰的基因型分型结果,扩增片段均小于125bp,分别检出7,5,8个等位基因和12,10,16种基因型,基因型分布均符合Hardy-W e inberg平衡。3个基因座在河北汉族人群的非父排除率和个人识别力分别为0.3530、0.3637、0.4901和0.7954、0.8113、0.8913。结论3个m in iSTR基因座在DNA高度降解检材的法医学分析中具有较高应用价值,并且在河北地区汉族人群中具有较好的遗传多态性。 Objective To study the value of application in samples with DNA degraded for three miniSTR loci D1S1677, D4S2364 and D10S1248,and investigate the genetic polymorphism of the three miniSTR loci in Han population from Hebei. Methods Three loci were amplified by PCR. The PCR products were separated by an ABI3100 Avant Genetic Analyzer and the results were analyzed using GeneScan 3.7 and Genotyper 3.7 software. Results Each locus was successfully genotyped with the length of the product less than 125bp. A total of 9 degraded samples were well genotyped by miniSTR technique. Among the 115 individuals investigated, 7, 5 and 8 alleles, and 12, 10 and 15 genotypes were detected at D1S1677, D4S2364 and D10S1248 respectively. No deviation from Hardy-Weinberg equilibrium was observed in these loci. The excluding probability of paternity and the discrimination power were 0. 3530, 0. 3637and 0.4901, and 0.7954, 0.8113 and 0.8913 for D1S1677, D4S2364 and D10S1248 respectively. Conclusion The three miniSTR loci were of benefit for the forensic analysis for degraded samples and have good polymorphism in Chinese Han population in Hebei.
