摘要
采用Langendorff灌注模型,对心肌缺血/再灌注损伤中氧自由基清除酶系统超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GPx)和乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK)、丙二醛(MDA)、超微结构的变化及硒的抗氧化效应进行了观察。随缺血时间的延长,LDH、CK活性及MDA的含量增加,而SOD、GPX的活性降低,此时超微结构损伤严重。预先给St·Thoma’s停搏液中加入亚硒醚钠再进行缺血/再灌注,冠脉流出液中的LDH、CK活性降低,心肌MDA含量减少;而心肌SOD、GPX的活性明显高于单纯缺血/再灌注时,超微结构损伤亦较轻。因此,说明了含硒St·Thoma’s停搏液对大鼠心肌缺血/再灌注的损伤具有保护作用。
We adopted the model of langendorff perfusion in order to observe superoxide dismutase(SOD),Glutathione peroxidase(GPX), Lactate dehydrogenase(LDH), Cretine phosphokinase (CK),malondialdehyde (MDA)and ultrastructure of oxygen free radicals(OFR)cleaning enzymes during ischemia reperfusion of myocardial injury.With time of ischemia prolonging,the contents of LDH,CKand MDA increased,and the activities of SOD and GPX decreased, at the same time, heavy damage of ultrastructure was also observed. But if selenium was added to St. Thoma's cardioplegic solution first,then, ischemia referfusion,the concentration of LDH and CK in the coronary vessel's sinus veflux solution,and the contents of MDA were lower,and the activities of SOD and GPX in mycardical tissue were significantly higher than those in using St.Thoma's Cardioplegic Solution, and the damage of ultrastructure was also slight.This suggests that St. Thoma's Cardioplegic Solution has the protective effect on rats' myocardial ischemia and reperfusion injury.
