摘要
用患有进展期S180肉瘤的小鼠进行的实验证实,多抗乙素(PB)对小鼠S180肉瘤有明显的抑制作用,使肿瘤生长延缓,坏死显著增加,瘤周瘤内淋巴细胞、巨噬细胞浸润明显增多,荷瘤鼠脾重明显增加,脾白髓动脉周围淋巴鞘及生发中心均增大,同时表明经PB治疗的肿瘤组织显示明显的出血坏死,坏死周边部瘤组织明显充血水肿,有更多的中性粒细胞浸润及小血管内血栓形成。封闭巨噬细胞后PB的抑瘤作用消失。结合以前的研究说明:PB的抑瘤作用主要是通过活化的免疫活性细胞(巨噬细胞、T细胞等)产生的TNF等细胞活性物质实现的。PB与阿霉素或5-氟尿嘧啶联用后,在抑瘤作用、肿瘤坏死比率、瘤周瘤内免疫活性细胞浸润及脾重增加等方面明显优于其单纯应用,证明它们之间存在明显的协同作用,且PB能明显减轻阿霉素、5-氟尿嘧啶对骨髓的抑制作用。PB+ADM组与PB组或ADM组相比,P<0.01;PB+5-Fu组与PB组或5-Fu组相比,P<0.01。从表1可知,PB、ADM、5-Fu对小鼠S180肉瘤均具有显著的抑制作用,ADM或5-Fu与PB联用后,抑瘤作用明显优于ADM或5-Fu或PB单用,且PB与5-Fu联用后有2例皮下肿瘤消失。2用药第4天各组月中?
our repeated experiments on mice suffering from advanced S180 Sarcoma showed that polyactin B could strongly suppress the growth of the tumor,increase the ratio of tumor necrosis area (TNA) to tumor total area(TTA) and the infiltration of lymphocytes, macrophages in the periphery of tumor and within the tumor, enlarge the spleen of tumor-bearing mice, especially the periarterial lymphatic sheath and the follices. In addition, there was more prominant hemorrhage, necrosis,and strongly hyperemia, edema, neutrophil infiltration and micro-thrombosis around the necrotic areas in the tumors of PB treated group.The tumor-sup-pressive activity of PB almost totally disappeared after blocking MΦsystem.Combined with our previous study, it is indicated that TNF produced by PB activated immunocompetent cells is the major mediator in PB mediated tumor-suppression; when Adriamycin or 5-Fluorouracil was used with PB,the tumor suppression effect of them would be enhanced and the side-and toxic effect was lightened.
