脐血浆内源性类洋地黄物质的部分纯化与鉴定 被引量:2

摘要 选用C18预处理柱分离脐血浆内源性类洋地黄物质(Endogenousdigitalis-likesubstance,EDLS),应用高效液相色谱分析法(HPLC)对其进行部分纯化,观察部分纯化的EDLS对红细胞摄取86Rb的抑制作用,并比较其与哇巴因在高效液相色谱图上的特点。结果表明:脐血浆第二份HPLC洗脱液(滞留时间4~6min)对红细胞摄取86Rb具有强烈的抑制作用;其抑制率为(89±3)%,明显高于其它洗脱物份别及孕妇、健康成人对照组血浆第二份洗脱物的抑制率[分别为(73±5)%和(43±3)%,P值均<0.01];部分纯化的EDLS、哇巴因及两者混合物的滞留时间基本一致,提示EDLS与哇巴因的结构相似。 Plasmas from cords, pregnant women and healthy volunteers were used. High performance liquid chromatography(HPLC) was employed to analyse and partially purify the isolated EDLS on C18 disposable columns. Inhibition of the purified EDLS on 86Rb uptake by intact erythrocytes was observed. The chromatographic retentions of EDLS and ouabain were compared by analytical HPLC using a C18 reversed phase column. The results were as follows: (1) Fraction 2 strongly inhibited 86Rb uptake by intact erythrocytes; (2) The inhibition rate of Fraction 2 from cord plasma was(89±3)%,Which was obviously higher than those of the other fractions in the same sample and the Fraction 2 from pregnant and the normal groups [(73±5)% and (43±3)%, respectively] (all P<0.01); (3)The retentions of EDLS, ouabain and the mixture of both were almost equal. It suggests that EDLS is similar to ouabain in structure.
