血塞通治疗大鼠急性局灶性脑缺血的研究 被引量:4

摘要 目的:观察血塞通对大鼠急性局灶性脑缺血的治疗效果。方法:以32只SD大鼠大脑中动脉阻塞(MCAO)持续脑缺血模型为研究对象,分为假手术组、模型组、尼莫地平组、血塞通组共4组,分析其术后4 h、24 h、72 h的神经功能缺损评分及行为学评分,比较各组的脑梗死体积,用分光光度法检测Na+,K+-ATPase、Ca2+-ATPase、Mg2+-ATPase的活性,用HPLC法检测ATP、ADP、AMP、TAN的含量。结果:(1)各治疗组梗死灶体积明显小于模型组(P<0.01)。(2)除假手术组外,各组动物从4 h至72 h神经功能缺损评分及行为学评分均有所降低,72 h时尼莫地平组、血塞通组与模型组比较,差异均有统计学意义(P<0.01),各治疗组之间差异无统计学意义(P>0.05)。(3)假手术组、治疗组Na+,K+-ATPase,Mg2+-ATPase活性均较模型组高(P<0.05);血塞通组Na+,K+-ATPase较尼莫地平组高(P<0.01)。(4)各组AMP,ADP差异无统计学意义(P>0.05);模型组ATP均较假手术组低(P<0.01),血塞通组、尼莫地平组较模型组高(P<0.05)。模型组、尼莫地平组TAN较假手术组低(P<0.05),血塞通组与假手术组比较,差异无统计学意义(P>0.05)。结论:血塞通能降低急性脑缺血大鼠神经功能评分,减轻脑梗死体积,改善缺血区脑组织能量代谢,效果略优于尼莫地平。 Objectives: To observe the therapeutic effect of Xuesaitong (ip) on acute cerebral infarcts. Methods: Thirty - two Sprague- Dawley (SD) rats were randomly divided into 4 groups: fake surgery group, model group, nimodipine group, Xuesaitong group. The neural function status deficiency and cerebral infarcts volume were observed. The activities of Na^+ , K^+ - ATPase,Ca^2+ - ATPase, Mg^2+ -ATPase were analysed by spectro photometry. The levels ATP, ADP, AMP and TAN were detected with high -pergomance liquid chromatography. Results : ( 1 ) The cerebral infarcts volumes of all the therapeutic groups were lower than that of the model group ( P 〈0.01 ). (2) Except the fake surgery group, the neural function grades and the behavior grades of all the groups were reduced from 4 h to 72 h. The nimodipine group and the Xuesaitong group had statistical significance than the model group at 72 h (P 〈 0.01 ). All the therapeutic groups had no statistical significance ( P 〉 0.05 ). (3) The activities of Na^+ , K^+ - ATPase, Mg^2+ - ATPase of the fake surgery group and the therapeutic groups were higher than the model group (P 〈 0.05 ). The activities of Na^+ , K^+ - ATPase of the Xuesaitong group were higher than the nomodipine group( P 〈 0.01 ). (4) There is no statistical significance among all the groups (P 〉 0.05 ). The ATP levels of the model group were lower than the fake surgery group (P 〈 0.01 ). The nimodipine group and the Xuesaitong group were higher than the model group ( P 〈 0.05 ). The TAN of the model group and the nimodipine group were lower than those of the fake surgery group (P 〈 0.05 ). The Xuesaitong group has no statistical significance than the fake surgery group ( P 〉 0.05 ). Conclusion : Xuesaitong can effective protect nerve cells, promote ischemic brain organize repair and improve energy metabolism. Its effect is prior to nimodipine.
