摘要 本论文以魔芋、番鬼芋、土壤、海水等从自然界采集到的样品为实验材料,经过富集培养、分离纯化、选择性平板分离,筛选出产β-甘露聚糖酶的菌株,再经过平板水解圈初筛得到20株酶活较高的菌株,经过摇瓶复筛,选出一株酶活最高的菌株,其酶活为15.66U/ml。对该菌株进行60Co诱变,经过分离、平板水解圈初筛和摇瓶复筛,得到一株酶活为31.89U/ml的菌株,其酶活是出发菌株的2.04倍。 By using the methods of enriching culture, isolation and selective plate culture, β-mannanase producing strains were isolated from konjak, spirit taro, soil and sea water, which were gathered from nature. 20 strains of high enzyme activity were got after the first screening by selective plates. Then the strain with the highest enzyme activity of 15.66U/ml was got after the second screening by shaking culture. To enhance the enzyme activity, mutagenesis of 60^Co was adopted. Then the screening methods were used again and one strain with the highest enzyme activity of 31,89U/ml, 2.04 times as its parent strain, was obtained.
