摘要
通过响应面分析法与旋转正交回归法对鹿茸多糖的提取工艺进行优化,以期得到最佳工艺。应用响应面分析法对酶解温度、时间、pH值进行考察,得到最佳工艺参数为:温度54℃,时间11h,pH8;确定醇析最适浓度为70%;对三氯乙酸除蛋白法、单宁除蛋白法及酶-Sevag除蛋白法进行比较,并采用旋转正交回归法对酶-Sevag法进行分析,证明当多糖溶液:Sevag试剂=1:1,氯仿:正丁醇=4.2:1时,除蛋白效果最佳。应用最佳工艺后,多糖提取率比传统方法提高了48.35%。
Extraction Process of Pilose antler polysaccharides (PAPS) was optimized by response surface analysis and orthogonal method. The results showed that the optimum temperature, the reaction time, the pH value and the ethanol concentration of enzyme hydrolysis were 54℃, 11 h, 8 and 70%, respectively. Comparing the effects of deproteinization by Trichloroacetic acid and Tannin with deproteinization by enzyme and investigating the Enzyme-Sevag methods by orthogonal design, the method of enzyme-sevag was the best. When the ratio ofpolysaccharide and sevag solution was I:1 and Trichloromethane / n-butanol (V/V) was 4.2:1, the deproteinization effect of sevag method was the best. The productivity of PAPS improved 48.35% as compared with traditional method.
