摘要
对一株嗜碱芽孢杆菌产环糊精糖基转移酶的发酵条件进行了研究。利用单因素试验和响应曲面试验获得该菌株产环糊精糖基转移酶的最佳条件为:接种量3%;培养温度30℃;pH10.5;发酵培养基的组成为玉米粉2%,酵母膏1.5%,玉米浆5%;250ml三角瓶装液量为30ml;270rpm振荡培养3d,其发酵液酶活可达5400U/ml左右。10L罐发酵时酶活可达5820U/ml。
The fermentation conditions of cyclodextrin glycosyltransferase (CGTase) produced by Bacillus alkalophilus 12- 7 were studied. Optimal fermentation conditions were determined by single factor experiment and response surface methodology (RSM), and the optimal conditions were as follows: amount of inoculation is 3%, incubation temperature is 30℃, pH value is 10.5; and the compositions of fermentation media are 2% maize powder, 1.5% yeast extract and 5% corn steep liquor; and load of 250ml flask is 30ml, shaking incubation is 3 days at 270rpm and the enzyme activity could reach 5400 U/ml. Enzyme activity couhl reach 5820U/ml in 10L termentor.
