高效液相色谱法比较不同种属和产地罗布麻叶中金丝桃苷的含量

Comparison of hyperoside in folium apocyni veneti of different species and areas by HPLC

目的建立罗布麻叶中有效成分金丝桃苷的HPLC含量测定方法，并且对不同种属和产地的罗布麻叶进行比较。方法采用Shim-Pack VP-ODS色谱柱；乙腈-四氢呋喃-0．1％醋酸（15．5：5．5：79）为流动相；流速1．0mL·min^-1；检测波长为360nm；柱温为30℃；进样量20μL。结果金丝桃苷线性范围0．01～0．2mg·mL^-1（r=0．9999）；重复性试验RSD=1．02％（n=5）；平均回收率为101．5％，RSD=2．1％；罗布麻属罗布麻叶中金丝桃苷含量在0．60～0．91％，白麻属两种含量在0．39～0．55％。结论本方法结果准确，重现性好，发现罗布麻属和白麻属两属之间金丝桃苷含量差异明显，为建立完善罗布麻叶质量标准提供依据。

OBJECTIVE To establish a method for determination of hyperoside in Luobuma by RP-HPLC and compare the difference between samples from different resources. METHODS The assay was performed on Shim-Pack VP-ODS eolumn, acetonitrile-tetrahydrofuran-0.1% acetic acid （15.5 ： 5.5 ： 79） was used as mobile phase, the rate of flow was 1.0 mL/min, detection wavelength was 365 nm, temperature of column was 30℃. RESULTS There was a good liner relationship within the concentration range of 0.01～0.2 mg·mL^-1 （r = 0.999 9）. The average recovery of hyperoside was 101.5 % and RSD was 2.1% （n = 6）. The content of hyperoside was 0.60～0.91% in samples from Apoeynum linn, and 0.39～0.55% in those from Poaeynum baill. CONCLUSION The content of hyperoside in two species of Luobuma was found to be different. The method developed is reliable and accurate and can be used for quality control of folium apocyni veneti.
