清醒大鼠膀胱测压建立梗阻性逼尿肌不稳定模型

摘要 目的探讨通过清醒膀胱测压建立梗阻性逼尿肌不稳定(detrusor instability,DI)模型的方法。方法成年雌性Wistar大鼠于会阴部行不完全性尿道结扎制作尿道出口梗阻模型,6周后耻骨上膀胱造瘘,于清醒状态下行充盈性膀胱测压,观察膀胱容量、压力变化、排尿反射及逼尿肌不稳定的发生情况。结果大鼠清醒测压时保持排尿反射,梗阻组发生逼尿肌不稳定的占76.17%,单位时间发生率(1.38±0.80)次/min,逼尿肌不稳定组膀胱最大压力(93.62±12.91)cmH2O和容量(1.20±0.41)mL较对照组(51.50±8.78)cmH2O(、0.33±0.08)mL显著增高(P<0.01)。结论清醒大鼠膀胱测压是建立梗阻性逼尿肌不稳定模型的可靠和实用的方法。 Objective To investigate the methods of construct detrusor instability (D1) animal model by in vivo cystometric evaluation in conscious rats. Methods Bladder outlet obstruction was created in female Wistar rats by partially ligating the proximal urethra in the perineum for 6 weeks. Cystometries were performed in conscious rats after suprapubic bladder catheter implantation. The bladder capacity and pressure were measured and the incidence of detrusor instability was studied. Results Reflex micturition remained in conscious rats during cystometric evaluation. The incidence of D1 developed in obstructed rats was 76.17% . The mean frequency is (1.38 ± 0.80)/rain. Bladder pressure and capacity increased from (51.50 ± 8.78) cmH20 and (0.33 ± 0.08) mL in control rats to (93.62 ± 12.91 ) cmH20 and ( 1.20 ± 0.41 ) mL in obstructed rats respectively ( P 〈 0.01 ). Conclusions This study suggested that cystometric evaluation in conscious rats is reliable and reproducible for the construction of detrusor instability animal model.
