摘要
目的探讨益心酮(Yixintong,YXT)注射液对大鼠实验性脑出血(intracerebral hemorrhage,ICH)的保护作用。方法应用立体定位技术,自大鼠尾动脉取不抗凝血液50μl缓慢注入尾状核,建立大鼠实验性脑出血模型,观察其脑出血后脑水肿的变化规律和脑水肿对其行为学的影响,并对脑组织进行病理学观察,检测大鼠血浆及脑组织匀浆中超氧化物歧化酶(SOD)活性、丙二醛(MDA)及一氧化氮(NO)的含量。结果益心酮注射液可明显降低出血脑组织的脑水分含量,明显改善神经行为,减轻脑组织病理改变,并可升高血浆及脑组织匀浆中SOD活性,降低血浆及脑组织匀浆中MDA及NO含量(P<0.05或P<0.01)。结论益心酮注射液可减轻脑出血后神经细胞损伤、降低脑水肿、改善神经行为,其机制可能与抗自由基、降低NO毒性有关。
Objective To investigate the protective effects of Yixintong(YXT) injections on experimental intracerebral hemorrhage (ICH) in rats. Methods Rat ICH models were established by injecting autologous blood into the caudate nucleus of rats stereotaxically. The changes of brain edema after ICH and the effects of brain edema on neurogenic behavior were observed. For brain tissues, histopathological tests were performed. The levels of superoxide dismutase(SOD), malondialdehyde(MDA) and nitric oxide(NO) in plasma and brain tissues were measured. Results After rats were treated with YXT injections, rain water contents in hemorrhagic tissues were significantly decreased, and neurogenic behaviors were significantly improved, and pathological changes of brain tissues subjected to ICH were significantly relieved, In the YXT-treated groups, in plasma and brain tissues of rats, the levels of SOD were increased, and the contents of MDA and NO were decreased ( P 〈 0.05 or P 〈 0.01 ). Conclusion Yixintong injections can improve neural cells lesions, brain edema, and durogenic behavior after ICH, The mechanism of protective effects may be associated with anti free radicals and suppressing NO.
