川芎嗪对大鼠胶原性关节炎滑膜VEGF及VEGFmRNA表达的影响 被引量:20

Effects of tetramethylpyrazine on the expression of vascular endothelial growth factor in synovium of collagen-induced arthritis in rats

摘要 目的观察川芎嗪对大鼠胶原性关节炎滑膜血管生成及VEGF表达的影响。方法Ⅱ型胶原皮内注射诱导W ist-ar大鼠关节炎,给予不同剂量的川芎嗪连续灌胃30 d。检测关节炎指数积分和炎症足垫厚度,免疫组化计算滑膜微血管密度,W estern B lot和RT-PCR方法分别检测滑膜血管内皮生长因子蛋白和mRNA的表达。结果100 mg.kg-1.d-1川芎嗪组的大鼠关节炎指数和足垫厚度均低于生理盐水组(P<0.05);滑膜微血管密度降低,VEGFmRNA表达及VEGF蛋白表达也明显减少,与模型组相比差异均有显著性(P<0.05)。结论川芎嗪可减轻大鼠胶原性关节炎的发展,抑制滑膜的血管病理性增生,其机制与抑制滑膜表达血管内皮生长因子密切相关。 Aim To explore anti-angiogenic effect of tetramethylpyrazine (TMP) in the synovium of collagen-induced arthritis (CIA) in rats. Methods CIA rats were treated with different doses of TMP. The effects of treatment were monitored by arthritis index, footpad thickness, microvessels density as well as the expression of vascular endothelial growth factor (VEGF) protein and mRNA in the synovium. Results Compared with CIA models, 100 mg · kg^-6·d^-1 TMP remarkably reduce the arthritis index, footpad thickness, microvessels density, and the expression of VEGF protein and mRNA in the synovium (P 〈 0. 05 or P 〈 0. 01 ). Conclusions TMP can attenuate the degree of chronic inflammation and angiogenesis in the synovium of CIA rats, and its mechanism might be associated with inhibiting the expression of VEGF.
