不同转移潜能的人前列腺癌细胞原位转移模型的建立与比较 被引量:1

摘要 目的建立不同转移潜能的人前列腺癌细胞转移模型,并比较其结果。方法利用人前列腺癌细胞系PC3单克隆细胞株裸鼠皮下移植瘤,通过外科原位移植技术植入BALB/c-nu/nu雄性裸小鼠,建立人前列腺癌高、低转移细胞株转移模型;并观察比较两细胞株转移模型的原位成瘤率、自发转移率、两细胞株形态学特征和转移抑制基因CD44s的表达。结果人前列腺癌高转移细胞株裸鼠原位种植后原位成瘤率100%,主动脉旁淋巴结转移率为100%,低转移细胞株裸鼠原位种植后原位成瘤率100%,主动脉旁淋巴结转移率为40%,两者主动脉旁淋巴结转移率差异有显著性(P<0·05)。高、低转移细胞株形态学特征亦不同。CD44s蛋白在人前列腺癌高转移性单克隆细胞株PC3-H中的表达强度低于低转移性单克隆细胞株PC3-L。结论遗传背景相似的人前列腺癌高、低转移细胞株原位移植转移模型为研究人前列腺癌转移机制提供了有力工具。 Objective To establish metastatic models of human prostate cancer cell clones with different metastatic potential and compare their different results. Methods In this study histologically intact tumor tissues of human prostate cancer cell clones( derived from prostate cancer cell line PC3 ) harvested from subcutaneous tumor in athymic nude mice were implanted orthotopically in nude mice(BALB/c-nu/nu) ,thus the metastatic models of human prostate cancer cell clones with high and low metastatic potential were established. The take rate, spontaneous metastatic rate, morphological characteristics of pathology and expression of tumor metastasis suppressor gene CD44s were detected and compared. Results The take rate and periaortic lymph node metastatic rate of orthotopic implantation of high metastatic potential cell clone were all 100% ; while the take rate and periaortic lymph node metastatic rate of orthotopic implantation of low metastatic potential cell clone were 100% and 40% respectively. And there was significant difference between them in periaortic lymph node metastatic rate ( P 〈 0. 05 ). The morphological characteristics of human prostate cancer cell clones with high and low metastatic potential were different from each other. We observed lower expression of CD44s in high metastatic potential cell clone. Conclusion The metastatic models implanted orthotopically with human prostate cancer cell clones with different metastatic potential, which have similar genetic background , provide useful models for investigating the mechanisms of human prostate cancer metastasis.
