VEGF和八因子相关抗原在人脑胶质瘤中的表达及意义 被引量:1

摘要 目的通过检测人脑胶质瘤细胞中血管内皮生长因子(VEGF)和八因子相关抗原(Ⅷ-R Ag)的表达,探讨VEGF在脑胶质瘤发生和发展中的作用。方法正常脑组织对照标本3例,取自脑外伤后内减压的病人。62例胶质瘤病人的手术标本,其中胶质母细胞瘤23例,间变胶质细胞瘤20例和星形胶质细胞瘤19例。采用VEGF多克隆抗体、兔抗人八因子相关抗原(Ⅷ-R Ag)行常规S-P免疫组化染色,观察并计算VEGF免疫活性指数和微血管密度值。结果VEGF染色指数随着肿瘤恶性度的升高而增高;脑胶质瘤组织微血管密度随着肿瘤级别的升高而增高;脑肿瘤病理分级和VEGF(r=0.941)的表达及与MVD(r=0.907)之间呈正相关性。结论VEGF可能通过促进肿瘤组织内血管生成,在人脑胶质瘤的形成与发展过程中起着重要作用,其表达水平对判断恶性程度及预后有指导意义。 Objective To explore the relationship between the grade of glioma and the expression of VEGF and Ⅷ-R Ag by detecfion the expression of VEGF and Ⅷ-R Ag in human gliomas and normal brain tissues.Methods Three normal brain tissues were obtained from the other patients as the control group. Sixty-two tissue specimens were obtained from patients with human brain glioma. The tumors were diagnosed as low-grade astrocytoma in 19 cases, anaplastic astrocytoma in 20 cases, and glioblastoma in 23 cases according to the World Health Organization criteria (1990). Immunohistochemical staining of paraffin tissue sections of tumor and normal brain specimens were used for detection the expression of VEGF and Ⅷ-R Ag. The immunoreactive score of VEGF and Microvessel Density (MVD) were calculated. Results The expression of VEGF and MVD increased with the grade of glioma, and significant diference was found among the different grade of glioma; MVD increased, with the grade of glioma,and significant diference was also found among all grades; The malignant grade of glioma was positively correlated to the expression of VEGF( r = 0.941 )and MVD( r = 0.907). Conclusion Tiffs evidence suggests that VEGF play important role in forming and developing of human brain glioma by stimulation the Angiopoiesis. VEGF expression may reflect the proliferating activity of the tumor cell and prognosis in brain glioma.
