利用SELDI-TOF-MS技术从结直肠癌患者血清中筛选特异的生物标记物

摘要 目的通过对血清中蛋白质组的质谱分析来发现可用于结直肠癌鉴定的生物标记物。方法随机取73例结直肠癌患者血清、31例正常人血清及16例结直肠相关良性疾病患者血清。将血清与金属亲和表面(IMAC3)芯片结合后,用蛋白芯片仪读取数据并进行软件分析。结果软件分析结果显示3类血清蛋白质在质荷比为4 467,8 131,8 939,9 192,9 134,8 221,5 928,8 324,11 732 D 9处含量有显著差异,得到树状分类规则,其分类准确率为98.33%(118/120),灵敏度为97.26%(71/73),特异性为100%(47/47)。结论用SELDI-TOF-MS技术对正常人、良性病患者及结直肠癌患者进行比较蛋白质组学研究,发现了结直肠癌病理发展中生物标记物。 Objective This study is aimed at discovering biomarkers to diagnose colorectal cancer. Methods Serum samples comprises 73 cases of colorectal cancer patients, 31 healthy people, and 16 cases of benign colorectal polypus. The sera were combined with the surface of the IMAC3 proteinchip. Then reading data of SELDI-TOF-MS was analyzed by software. Results At the key M/Z values of 4 467, 8 131, 8 939, 9 192, 9 134, 8 221, 5 928, 8 324, 11 732 D, protein contents of the three classes in preliminary group were obviously different by the software analysis. And the classification rule of tree was discovered, and corresponding correct ratio was 98.33 % ( 118/120), corresponding sensitivity was 97.26 % (71/73) and corresponding specificity was 100% (47/47). Conclusion Via comparative proteomics research in the sera of colorectal cancer patients and benign colorectal polypus and healthy people, biomarkers which can be used to diagnose colorectal cancer are discovered.
