摘要 目的观察高+Gx过载对猴肾脏组织c-fos表达的影响。方法9只雄性恒河猴,随机分为4组。对照组与实验1、2、3组经受+Gx及其作用时间分别为1 Gx/300 s、+15 Gx/200 s、+18 Gx/165 s和+21 Gx/140 s。使用专用动物离心机,过载暴露后的动物即刻处死、取材、制片、染色。用免疫组织化学方法观察猴肾脏组织c-fos表达。结果实验组猴肾脏的肾小球充血、肾小管浊肿。肾小管上皮细胞损伤程度呈现出较明显的G值量-效关系。对照组猴肾脏组织结构基本正常。各组动物肾小管上皮细胞有不同程度表达c-fos蛋白。对照组部分肾小管上皮细胞胞浆呈淡黄色,为c-fos弱阳性表达;实验组c-fos表达明显增强,阳性产物呈棕黄色颗粒状弥漫分布于肾近曲小管上皮细胞胞浆和胞膜,尤其近管腔面染色加深。实验3组(21 Gx/140 s)较实验1、2组c-fos表达有增强趋势。结论高+Gx过载作用可诱导肾脏组织中c-fos表达,其表达的强度随着+Gx值的增加有所增强。 Objective To observe the changes of c-fos expression in kidney tissue of rhesus macaques induced by +Gx loads. Methods Nine male rhesus macaques were randomly divided into four groups: control group (n=2) was exposed to +1 Gx/300 s, and experimental groups 1(n=2), 2 (n=2) and 3(n=3) were exposed to +15 Gx/200 s, +21 Gx/165 s and +21 Gx/140 s respectively. Renal tissue was fixed with 10 %buffered formaldehyde and embedded with paraffin, and histopathological changes were observed with light microscope. Immunohistochemical staining (S-P method) was used to detect the expression of c-fos in sections of kidney tissue. Results In the experimental groups, glomerular congestion and cloudy swelling of the tubular epithelial cells were found in the renal tissue. The degree of damages was significantly correlated with the increase of +Gx loads. There was no obvious pathological change in the control group. Immunohistochemical staining showed that expression of c-fos group was weak positive (light yellow) in control group and the intensity of staining in epithelial cells of renal tubules was enhanced in 3 experimental groups in a dosage-dependant manner. Conclusions Sustaind high +Gx exposure can induce the over-expression of c-fos in the kidney tissue of the experimental monkeys. There is a significant correlation of the enhancement of c-fos expression with the level of +Gx load.
