农杆菌介导法获得籼稻温敏核雄性不育系转Bt基因植株 被引量:6

Production of Bt-transgenic Plants Indica Thermo-sensitive Genic Male Sterile(TGMS) Rice Line Using Agrobacterium-mediated Method

摘要 通过农杆菌介导法将Bt融合型基因cry1Ab/cry1Ac导入籼稻温敏核雄性不育系03B198S和03B199S的愈伤组织,经过多轮潮霉素筛选,获得了一批独立转化体,同时,从每一个独立转化体至少再生出5株绿苗。经PCR、RT-PCR检测证实该Bt融合型基因已整合到部分受体基因组中,并表现出不同程度的表达。这些阳性转基因植株在异地异季低温条件下割蔸栽植,部分恢复可育,且表现出很强的抗螟虫性能。 The fused Bt gene crylAb/crylAc was introduced into the calli of indica “TGMS lines 03B198S and 03B199S using Agrobacterirum-mediated method. After multiple-cycle selections with hy gromycin,lots of independent transforrnants were obtained and at least 5 plants were regenerated from each transformant. The PCR and RT-PCR analysis of these transformants confirmed the integration, of the fused Bt gene into the genome of some recipient plants and the expression of its mRNA at different levels. Some positive transgenic plants after ratooning turned to be fertile at low temperature in the field of Hainan at winter,and showed highly resistant to leaf folders and stem borers.
