摘要 目的 建立组织样中氟尿嘧啶的药物浓度HPLC测定方法。方法 HPLC测定,按中国药典2005版规定做方法学验证。结果 方法的主要指标如下:检测限为0.017μg·g^-1,日内精密度(RSD)约3.0%,日间精密度(RSD)约7.1%。方法 的回收率为92%~98%。结论 被测组织样的测定结果在0.14~40.85μg·g^-1。之间。该方法符合2005年中国药典关于生物样品分析方法的规定,是检测组织样品中氟尿嘧啶的药物浓度的可行方法。 Aim To establish a HPLC method for the determination of 5-fluorouracil in tissue. Methods 5-Fluorouracil was determined by HPLC according to Chinese Pharmacopoeia published in 2005. Results The main parameters of this method were as follows : the limit of detection was 0. 017 μg · g^-1 ,the intra-day RSD was 3.0% ,the inter-day RSD was 7.1%. The recoveries were between 92% and 98.0%. Conclusion The method meets the criterion of China Pharmacopoeia 2005 for detecting biological specimen and is feasible for the determination of 5-fluorouracil in tissue.
