食管鳞癌中HIF-1α的表达与凋亡的相关性研究 被引量:2

摘要 目的探讨缺氧诱导因子1α(HIF-1α)和凋亡抑制因子(bcl-2)在食管鳞癌中的表达及二者之间的相关性。方法采用SP免疫组织化学染色法检测60例食管鳞癌组织和10例正常食管黏膜中HIF-1α蛋白和bcl-2蛋白的表达情况。结果HIF-1α在正常食管黏膜不表达,食管鳞癌中的阳性表达率为61.7%,HIF-1α蛋白的阳性表达率随食管癌体积增大、组织分化程度降低而增高(P<0.05),有淋巴结转移者的阳性表达率(78.3%)显著高于无淋巴结转移者(51.4%),(P<0.05);HIF-1α蛋白与bcl-2蛋白表达呈负相关(P<0.01)。结论HIF-1α蛋白的高表达可能促进了食管鳞癌癌细胞的增殖和组织分化程度的降低,并参与了淋巴结转移;它还可能通过调节凋亡抑制因子bcl-2的水平而发挥抗凋亡作用。 [Objective] To explore the expression of HIF-1α and the relationship between its expression and bcl-2 in esophageal squamous cell carcinoma. [Methods] Expression of HIF-1α and bcl-2 were assessed by immunohistochemistry staining in 60 cases of esophageal squamous cell carcinoma and 10 cases normal esophageal tissues. [Results] HIF-1α was hardly expressed in all normal mucosa epithelium, while its expression rate was 61.7% in 60 cases of esophageal squamous cell carcinoma. The positive expression rate of HIF-1α increased as the size increasing and the degree of differentiation decreased of the primary tumors (P 〈0.05), and its positive rate in the group with lymph node metastasis (78.3%) was much higher than that in the group without lymph node metastasis (51.4%, P 〈0.05). That the relationship between HIF-1α and bcl-2 was negative (P 〈0.01). [Conclusions] The over expression of HIF-1α may promote the proliferation and poorly differentiation of esophageal squamous cell carcinoma, and may take part in the metastasis of lymph node.
