摘要
目的研究在外源性细胞因子的作用下,蜕膜细胞对B细胞IgG分泌的影响,以探讨蜕膜局部免疫微环境的免疫学特点。方法在培养的蜕膜细胞中加入细胞因子-γ干扰素(Interferonγ,IFN-γ)、白细胞介素-2(Inter-leukine-1,IL-2)、白细胞介素-6(Interleukine-6,IL-6)和表皮生长因子(EpidermalGrowthFactor,EGF),用放射免疫法测定每分钟的脉冲指数(countperminute,cpm)来计算B淋巴细胞分泌的免疫球蛋白G(ImmunoglobulinG,IgG)的值。结果正常早孕蜕膜细胞的培养上清液可以刺激B淋巴细胞IgG的分泌;经外源性细胞因子作用后的蜕膜细胞培养上清液,B淋巴细胞分泌的IgG显著上升(P<0.05),尤其是第12h实验组的cpm上升有显著效应(P<0.05),24、48h的实验组cpm有逐渐下降趋势。结论在外源性细胞因子的作用下,蜕膜局部体液免疫反应增强,但这种增强可能是蜕膜局部免疫的自身调节作用,以维护妊娠的正常进行。
Objective To study the influence on early pregnancy decidual cells stimulated by exogenous cytokines on the secretion of IgG by B lymphocytes in vitro, and to investigate the features of the local immunological microenvironment of decidua. Method Exogenous cytokines interferon - γ, interleukin - 2, interleukin - 6 and epidermal growth factor to the decidual cells cultures, and harvest its supernatant, then co-culture with the human peripheral blood B lymphocytes, and assess the cpm of IgG secreted by B ceils with radio- immunological method. Results The decidual cells of normal early Pregnancy after stimulation with exogenous cytokines can stimulate the secretion of IgG by B lymphocytes. The supernatant of the decidual cells stimulated by exogenous cytokins showed the same function, but such function had no relation to the concen- tration and types of cytokines, time was the only factor concerned with. Conclusion Function of the exogenous cytokines can increase the homural- mediated immune function of the decidual immunological micro- environment, but such function may be self- mediated by the local immune micro- environment of the deciduas which function to support the normal pregnancy.
