摘要
目的优化金丝桃细胞株的悬浮培养条件,提高黄酮产量。方法研究激素组成、激素浓度、pH值、蔗糖浓度、培养温度、摇床转速、接种量等因素对金丝桃细胞生长量和总黄酮含量的影响,确立高产细胞系。结果培养基成分MS(Murashige and Skoog培养基)+10-5mol/L NAA+10-6mol/LBA,25℃,pH值5.7,蔗糖含量3%,摇床转速90 r/min,接种量60g/L,为最佳的金丝桃细胞悬浮培养条件。结论获得的金丝桃悬浮细胞系稳定、可靠,可作为工业化生产黄酮的种细胞。
Objective To develop the yield of flavonoids via the optimization of culture conditions of Hypericurn chinense L. Suspension Cells. Methods The effects of hormone component, hormone concentration, pH value, sucrose concentration, culture temperature, rotation speed and inoculation quantity on the accumulation of total flavonoids and growth of Hypericurn chinense L.suspension cells were studied and the high flavonoids-producing cell lines were also established. Results The best culture conditions of Hypericurn chinense L. Suspension Cells were as follows: Murashige and Skoog(MS) medium supplemented with 10^-5mol/L NAA and10^-6 mol/L BA, 25 ℃, pH 5.7, 30 g/L sucrose, 90 r/min of rotation speed and 60 g/L inoculation quantity. Conclusion The obtained Hypericurn chinense L. Suspension Cells are stable and credible and can be used as the strain cells in the industrial manufacture of flavonoids.
