摘要
目的:探讨前列腺素E1对梗阻性肾病中转化生长因子-β1和结缔组织生长因子表达的影响。方法:60只Wistar大鼠,随机分为PGE1治疗组、生理盐水组和假手术组,然后制作梗阻性肾病动物模型,最后在光镜下观察肾脏组织的病理学改变;采用免疫组化法检测肾脏的TGF-β1和CTGF的表达,分析肾脏纤维化与TGF-β1和CTGF的表达之间的相关性。结果:①生理盐水组和PGE1治疗组大鼠的间质纤维化的程度均高于假手术组(P<0.05),大鼠肾组织TGF-β1和CTGF表达高于假手术组(P<0.05),且与Masson染色结果呈正相关关系;②PGE1组以上各项指标均低于生理盐水组。结论:PGE1可以抑制TGF-β1、CTGF的表达,减轻肾间质纤维化,从而防治梗阻性肾病。
Objective:To study the effect of prostaglandin Et (PGEL)on the expressions of transforming growth factor-β1 (TGF-β1) and connective tissue growth factor (CTGF) in tubulointerstitium of rats with obstructive nephropathy. Methotis: 60 Wistar rats were randomized into PGEt treatment group, saline group and sham operation control group. Model rats were induced by ligating unilateral ureter. The morphopathological changes of tubulointerstitium were observed by light microscopy and the expressions of CTGF, TGF-β1 were detected by immunohistochemical staining. The measured data were evaluated by Mais-2000 image analysis system. Results: ①Compared with sham operation control group, the level of tubulointerstitial fibrosis and the expressions of CTGF, TGF-β1 were increased obviously in PGEt treatment group and saline group. And the expressions of CTGF and TGF-β1 was positively correlated with Masson staining. ②All indexes of PGEt treatment group were decreased obviously compared with those of saline group. Conclusion: PGEt could alleviate tubulointerstitial fibrosis through inhibiting the expressions of CTGF and TGF-β1 in tubulointerstitium.
