摘要
目的:研究特异性抑制HPVE6基因的表达对宫颈癌CaSki细胞的影响.方法:构建表达HPV-16E6基因siRNA重组质粒,体外转染CaSki细胞后检测E6mRNA表达变化及E6主要作用靶分子P53的蛋白变化;流式细胞术分析细胞周期.结果:特异RNAi表达质粒转染CaSki细胞后明显降低了E6mRNA水平,且P53蛋白水平增高.流式细胞仪测定发现细胞停滞在G0-G1期,细胞增殖受到抑制.结论:采用RNA干扰技术能沉默CaSki细胞中整合的HPV-16E6基因的表达,使细胞中抑癌蛋白P53水平增高,导致肿瘤细胞生长停滞.
AIM: To study the effect on CaSki cells by inhibiting human papillomavirus (HPV) 16 E6 gene expression in CaSki cell. METHODS: The vectors expressing siRNA of HPV-16 E6 gene were constructed and transfected into CaSki ceils. The expression of E6 mRNA was detected by RT-PCR and the expression of P53 protein by Westen blot. CaSki cell cycle was analyzed by flow cytometry. RESULTS: After transfection, the expression of HPV-16 E6 mRNA was reduced and the expression of P53 was up-regulated in CaSki cell line. The proliferation of CaSki cells were inhibited and the cells were arrested at G0-G1 stage. CONCLUSION: RNAi can inhibite the expression of HPV-16 E6 gene in CaSki cells, which up-regulates the expression of tumor suppression protein P53 and results in growth retardation of CaSki cells.
