摘要
目的:观察榄香烯乳对胃癌细胞株SGC7901人端粒酶反转录酶(hTERT)表达及其启动子的影响,以深入了解榄香烯乳的抗癌机制。方法:以榄香烯乳处理胃癌细胞,分别采用实时定量PCR和免疫荧光标记法检测癌细胞hTERT mRNA和蛋白水平的表达。采用脂质体转染法将携带hTERT启动子和报告基因的质粒转染至胃癌细胞株SGC7901细胞中,2 h后加入不同浓度的榄香烯乳,孵育48 h后,分别检测其报告基因萤虫素酶活性。结果:榄香烯乳处理后,胃癌SGC7901细胞hTERT mRNA和蛋白水平呈浓度和时间依赖性下调。榄香烯乳处理组萤虫素酶活性下降,且呈浓度依赖性。结论:榄香烯乳可能通过下调hTERT启动子活性,抑制hTERT表达,从而抑制胃癌细胞端粒酶活性。
Objective: To investigate the effect of elemen on expression and promoter of human telomerase reverse transcriptase(hTERT) in gastric cancer cell. Methods: After gastric cancer cell line SGC7901 were treated with different dose of elemen, mRNA and protein of hTERT were determined by real-time PCR and immunofluorescense labeling assay, respectively. The plasmid inserted hTERT promoter was transfected into SGC7901 cells by oligofecamine, and different doses of elemen were added into culture media 2 hours later. After 48 hours, luciferase activity of gastric cancer cell was determined. Results: The expression of mRNA and protein of hTERT of cells treated with elemen were down-regulated in dose-and time-dependent manner. The luciferase activity of cancer cells treated with elemen were inhibited in dose-dependent manner. Conclusion: Elemene may suppress telomerase activity through inhibiting expression of hTERT promoter of gastric cancer cell.
