摘要
用果胶酶和纤维素酶酶解黄花梨花药制备染色体标本,从大量减数分裂中期I分裂相中选择分散良好的染色体进行拍照和组型分析,建立核型图。根据核型识别目标1号染色体,应用显微操作系统成功进行单染色体分离。
Chromosome specimens were made from the anthers of pear (Pyrus pyrifolia) variety Huanghuali, dissociated with the mixture of pectase and cellulase. Its meiotic metaphase I karyotype was established, based on the lengths of the chromosomes. Chromosome 1 was identified and microdissected, using microglass needles under a micromanipulator and transferred into an Eppendorf tube for further study. Such procedure proved successful for the microdissection of the target chromosomes of P. pyrifolia.
