摘要
采用改良的CTAB法,以玉米的黄化苗叶片为材料提取玉米基因组DNA,提取的DNA经0.8%的琼脂糖凝胶电泳检测。结果表明:此法具有快速、高效、DNA质量好和纯度高等优点。对所构建的选育高直链淀粉玉米群体的双亲进行了SSR-PCR检测,得到了清晰的多态性SSR标记,为玉米分子标记辅助育种打下了基础。
By using improved method of CTAB, genomic DNA of' maize yellow seedling was extracted, then detected by 0.8% gelose gelatin electrophoresis in this paper. In order to detect the availability of DNA extraction approaches, the high amylose-extender maize parents were detected by SSR-PCR, too. The results showed that the improved method of CTAB had displayed many fine characteristics such as high efficiency, perfect quality and high purity, and ideal amplification bands were received. So the improved method of CTAB and SSR-PCR provided a foundation for the molecular marker-assisted maize breeding.
