摘要
目的通过检测低氧状态血管内皮细胞一氧化氮合酶(eNOS)及细胞间黏附因子(ICAM-1)表达的变化,探讨低氧对血管内皮细胞功能的影响。方法采用逆转录-聚合酶链式反应(RT-PCR)及Western blot测定低氧条件下不同时间点大鼠动脉内皮细胞中eNOS、ICAM-1 mRNA及蛋白质的表达变化情况。结果同常氧组相比,低氧1 h时eNOS、ICAM-1 mRNA及蛋白表达无变化,而6、12、24 h时eNOS、ICAM-1 mRNA及蛋白表达明显升高(P<0.05),但eNOS mRNA与ICAM-1 mRNA的表达变化在同一时间点并不一致。结论低氧状态下血管内皮细胞eNOS与ICAM-1表达升高,可能参与了低氧性肺血管疾病的发病机制。
Objective By detecting the change of gene expression of eNOS and ICAM-1 to investigate the influence of hypoxia on pulmonary artery endothelial cells (PAEC) function. Methods Under condition of hypoxia, RT-PCR and Westernblot were used to detect the expression of eNOS and ICAM-1 mRNA and protein in PAEC in rats at different time points. Resuits The gene expression of eNOS and ICAM-1 was unchanged in PAEC after 1 h of hypoxia (P〉0.05). After 6 h, the expression of eNOS and ICAM-1 mRNA and protein was significantly increased (P〈0.05), but the change of the two factors did not consistent with time. The expression of eNOS mRNA was significantly higher after 6 h of hypoxia and the expression of ICAM-1 mRNA was significantly higher after 24 h of hypoxia. Conclusion Under condition of hypoxia, the gene expression of eNOS and ICAM-1 in PAEC was up-regulated, which might contribute to the pathogenesis of lung disease induced by hypoxia.
