TGF-β_1、C-Fos在单基因遗传自然发病型糖尿病小鼠颌下腺的表达

摘要 目的:探讨db/db(单基因遗传自然发病型)自发性糖尿病小鼠颌下腺转化生长因子β1(TGF-β1)、C-Fos蛋白的表达情况。方法:分别取3、4、6、8、10月龄db/db糖尿病小鼠颌下腺(实验组)及相应月龄的db/+m正常小鼠颌下腺(对照组),每组5只小鼠。SP免疫组化染色观察颌下腺TGF-β1及C-Fos的表达情况。结果:随着糖尿病的进展,db/db糖尿病小鼠颌下腺TGF-β1阳性细胞数逐渐增加,明显高于同龄对照组(P<0.01);各月龄糖尿病小鼠颌下腺C-Fos阳性细胞数逐渐减少,明显低于同龄对照组(P<0.01)。结论:db/db糖尿病小鼠颌下腺TGF-β1的表达上调可能与糖尿病间质纤维增生密切相关,而C-Fos的低表达可能与颌下腺实质的萎缩性形态学变化密切相关。 Objective.To study the expression of transforming growth factor-β1 (TGF-β1) and C-Fos in submaxillary salivary gland of C57BL/ksj -db/db mutant diabetic mice. Methods: 3,4,6,8,10 month-old db/db mice were used and the age-matched db/+m mice were taken as control respectively, each monthage had 5 mice. SP immunochemical technique was used to observe the expression of TGF-β1 and C-Fos in submaxillary salivary gland. Results: In db/db diabetic mice, TGF-β1 positive cell numbers in submaxillary salivary gland was obviously increased, while the C-Fos positive cell numbers significantly decreased compared with the age-matched control animals (P〈0. 01). Conclusions. In db/db diabetic mice, the increased expression of TGF-β1 in submaxillary salivary gland may has close relations with the proliferating of interstitial fiber and the decreased expression of C-Fos may has close relations with the atrophying of submaxillary salivary gland.
