超抗原和肿瘤抗原修饰树突状细胞的活性研究 被引量:3

摘要 目的金黄色葡萄球菌肠毒素B(SEB)和肿瘤抗原修饰树突状细胞(dendritic cell,DC)后,活化的DC与淋巴细胞(lymphocyte cells,LC)共孵,观察DC激活的LC在体外对结肠癌细胞的排斥性杀伤作用,以评价抗原修饰的DC活性。方法外周血分离出LC、单个核细胞,细胞因子诱导DC扩增,DC活化LC,对同种异型肿瘤细胞排斥性杀伤实验。结果SEB+肿瘤抗原修饰后的DC具有显著的淋巴细胞活化作用。加SEB比单独使用肿瘤抗原修饰DC后的活化淋巴细胞的功能强(P<0.05)。结论SEB和肿瘤抗原联合修饰可以明显增强DC的活性(P<0.05)。 Objective SEB (staphylococcal enterotoxin B) and tumor antigen jointly modified the DC. Then activated DC incubated with LC. The effect of LC that was activated by DC, act on the allotype colon cancer cells at extraorgan was observed. Through it the activity of DC modified by antigen can be evaluated. Methods To abstract the lymph cells and mononuclearcells from peripheral blood, and induce and amplify the DC by cytekine. LC activated by DC carried on repellency killing experiment of allotype tumor cells. Results DC modified by tumor antigen and SEB jointly had conspicuous lymphocyte activation in our experiment: Co-effect of DC modified by t μ mor antigen and SEB jointly was stronger than DC modified by tumor antigen solo (P〈0.05). Conchusions The activity of DC modified by superantigen and tumor antigen can be enhanced conspicuously (P〈0.05).
