摘要
目的探讨建立舌鳞状细胞癌淋巴道转移动物模型的实验方法。方法采用裸鼠爪垫皮下注射舌鳞状细胞癌Tca8113细胞的方法建立舌鳞状细胞癌淋巴道转移裸鼠模型,4周后收集腹股沟、腋窝、腹腔等部位淋巴结,进行H-E染色与EMA、CK18免疫组化染色以及体外舌鳞状细胞癌细胞纯化培养等检测,判定裸鼠模型建立的成功与否。结果H-E染色与EMA、CK18免疫组化染色发现淋巴结有散在或灶性鳞状上皮癌细胞转移,体外纯化培养的细胞呈上皮样生长。在35只受试裸鼠中,23只建模成功,转移率为67.5%。结论裸鼠爪垫皮下注射可成功建立舌鳞状细胞癌淋巴道转移模型,为舌鳞状细胞癌淋巴道转移的机理研究、药物干预提供动物模型。
Objective To establish the nude mice model of lymphatic metastasis using tongue squamous cell carcinomal (TSCC) Tca8113 cell line. Methods The lymphatic metastasis nude mice models were established by injecting Tca8113 cells into the nail pad, lymph nodes were collected after 4 weeks. The detection include the HE strain, immunohistochemistry strain of EMA and CK18 proteins and the purification and culture of tongue squamous cell carcinoma cells in vitro. Results The lymphatic metastasis rate was 67.5% (23/35). There are EMA and CK18 proteins expression and lymphatic metastasis carcinomal cells in the lymph node of nude mice of TSCC metastasis model. In purification and culture, the cells of epithelium like growth were observed in vitro. Conclusion Injecting Tca8113 cells into the nail pad can establish the lymphatic metastasis model. The model was regarded as basis study of the lymphatic metastasis mechanisms and the research of anti-lymphatic metastasis drug.
