蝙蝠葛碱在体外对轻度修饰的脂蛋白诱导人单核细胞血小板源生长因子B蛋白量的影响

Effect of dauricine on production of platelet-derived growth factor B protein induced by minimally modified lipoprotein in human monocytes in vitro

从正常人血中分离单核细胞，分别加入２５ｍｇ·Ｌ－１脂蛋白（ＬＤＬ，ＶＬＤＬ）及经Ｃｕ２＋轻度氧化修饰脂蛋白（ＭＭ－ＬＤＬ，ＭＭ－ＶＬＤＬ）进行培养．应用免疫细胞化学技术观察蝙蝠葛碱对脂蛋白诱导单核细胞产生血小板源生长因子Ｂ（ＰＤＧＦ－Ｂ）蛋白的影响．结果表明，ＬＤＬ和ＶＬＤＬ对单核细胞ＰＤＧＦ－Ｂ量无明显影响，ＭＭ－ＬＤＬ，ＭＭ－ＶＬＤＬ明显增强单核细胞ＰＤＧＦ－Ｂ量，而蝙蝠葛碱对此有显著抑制作用．结果提示，蝙蝠葛碱抗动脉粥样硬化作用可能与抑制ＳＰＤＧＦ的作用有关．

Human monocytes were prepared and cultured with native LDL, VLDL and minimally modified LDL (MM-LDL), minimally modified VLDL (MM-VLDL) by 10 μmol·L -1 Cu 2+ . The effect of dauricine on the production of platelet- derived growth factor B (PDGF-B) protein induced by lipoprotein was observed in vitro immunocyto- chemically. The results indicated that native LDL and VLDL had no effect on the production of PDGF-B protein, but MM-LDL and MM-VLDL apparently stimulated the production of PDGF-B protein in monocytes, and the latter effect was markedly inhibited by dauricine. It suggests that the antiathero- sclerotic effect of dauricine may be implicated in its inhibitory effect on PDGF production.
