摘要
目的:观察体外不同氟浓度对人脐静脉血管内皮细胞的损伤作用。方法:体外培养人脐静脉内皮细胞株HUVEC-304,在培养液中加入不同浓度的氟化钠,共分9组,分别为正常对照组及加氟组(120、240、360、480、6007、208、40、960μmol/L),连续培养24 h后,细胞计数并观察细胞形态,MTT比色法检测细胞活性,并检测培养液中NOS、OD、MDA的含量。结果:①氟的浓度为120、240μmol/L时细胞数目明显增多,从600μmol/L开始,内皮细胞数量明显下降,细胞形态呈损伤改变;②氟的浓度在120、240μmol/L时,MTT测定细胞活性明显高于正常对照组,以240μmol/L浓度时最高;浓度在600以上时细胞活力明显低于正常对照组,差异均有统计学意义(P<0.01);③培养液中NO从240μmol/L时开始降低,和正常对照组差异显著,SOD从240μmol/L时开始下降,差异均有统计学意义(P<0.05),MDA的含量逐渐升高。结论:低浓度的氟对脐静脉血管内皮细胞有促进增生作用;高浓度有损伤作用,且随剂量的增高而增重。
Objective: To observe the lesion of human umbihcal vein endothehal ceils (HUVECs) induced by different doses of fluoride. Methods: HUVECs were cultured with fluoride in various concentration ( 120, 240, 360, 480, 600, 720, 840, 960 μmol/L), 24 hours later, the morphologic changes were observed, the cell activity was measured by MqT method, and the MDA, NO and SOD were determined. Results: The ceil increased when treated with fluoride of 120 μmol/L to 240 μmol/L, whereas decreased with fluoride of 600 μmol/L to 960 μmol/L, and the cells were injured. The ceil activity of the group of 120 μmol/L and 240 μmol/L significantly enhanced compared with the normal control, but that of the group of over 600 μmol/L markedly lowered compared with the normal control. NO and SOD decreased, but MDA increased in the group of over 240 μmol/L. Conclusions: Fluoride of low concentration can promote the ceil activity, but that of high concentration can injure the ceil, and the higher the concentration is, the heavier the iniury is.
