The distribution of (CA)_n polymorphism in endothelial nitric oxide synthase gene and its association with diabetic nephropathy in type 2 diabetic patients

摘要 目的探讨内皮一氧化氮合酶基因(CA)_n多态性在新加坡2型糖尿病患者中的分布及与糖尿病肾病的关系。方法258例2型糖尿病患者人选。从全血中提取DNA,然后进行多聚酶链反应、凝胶电泳及测序。结果在新加坡的中国人、马来西亚人和印度人中各有23、22、20种(CA)_n多态性,其基因型分布差异有统计学意义(P<0.01)。新加坡中国人(CA)_n的分布与欧洲白人差异有统计学意义。该基因多态性与糖尿病肾病无显著相关性。结论(CA)_n多态性在新加坡3个种族中的分布差异有统计学意义,而且与欧洲白人的分布差异也有统计学意义,但与糖尿病肾病无显著相关性。 Objective To examine the association of the distribution of (CA) o polymorphism in intron 13 of the endothelial nitric oxide synthase ( eNOS ) gene with diabetic nephropathy in type 2 diabetic patients in Singapore. Methods A total of 258 type 2 diabetic patients with more than 10 years duration ( 150 Chinese, 71 Malaysians and 37 Indians) from the National University Hospital of Singapore were studied. Polymorphism was examined by DNA extraction, PCR, electrophoresis and sequencing. Results In the study, 23, 22 and 20 genotypes were found in Chinese, Malaysians and Indians, respectively. The distribution of the (CA)n polymorphism showed significant differences among ethnic groups in Singapore ( P 〈 0.01 ). Its distribution was also significantly different between reported frequencies in European Caucasians and Singapore Chinese. However, there was no significant association between ( CA ) n polymorphism and diabetic nephropathy. Conclusion The distribution of the eNOS gene ( CA ) n polymorphism shows significant differences among Chinese, Malaysians and Indians in Singapore, and between Singaporeans and Caucasians. This polymorphism does not show any association of the ( CA)n polymorphism with diabetic nephropathy.
