摘要
目的检测大鼠胚胎心脏发育过程中IFRD1基因mRNA及蛋白的表达。方法取大鼠胚胎12、15、19 d心脏,应用RT-PCR、Western-blot、免疫组织化学技术检测胚胎大鼠心脏IFRD1基因mRNA及蛋白表达。结果在大鼠胚胎12、15、19 d心脏中,IFRD1基因mRNA及蛋白均有表达,且呈逐渐下调趋势;其蛋白主要表达于心房、心室、肌小梁等处心肌组织,而在心内膜垫、瓣膜、流出道、大血管内层等处未见表达。结论IFRD1基因的表达随大鼠胚胎心脏的生长发育逐渐下调,且特异表达于大鼠胚胎心脏心房、心室、肌小梁等处心肌组织,可能与心脏发育密切相关。
Objective To investigate the expression of interferon - related developmental regulator 1 ( IFRD1 ) gene during development of embryos hearts in rat. Method Rat hearts in embryonic 12^th, 15^th, 19^th day were included and the expression of IFRDI gene was detected by RT- PCR, Western - Blot and immunohistochemistry. Results IFRD1 gene was expressed in rat hearts in 12^th, 15^th, 19^th day. The expression decreased during development of rat hearts. IFRD1 gene was mainly expressed in the atria, ventricles and trabecular muscles, while not in atrial- ventricular valves, outflow tracks and large arteries. Conclusion IFRD1 gene is down - regulated during development of rat hearts and mainly expression in the atria, ventricles and trabecular muscles.
