异丙基肾上腺素致豚鼠慢性心力衰竭模型的建立 被引量:3

The establishment of the animal model of guinea-pig chronic heart failure induced by isoproterenol

摘要 目的探讨药物诱导豚鼠慢性充血性心力衰竭(CHF)动物模型的制作方法。方法采用皮下注射异丙基肾上腺素(1次/d)建立豚鼠心肌缺血型CHF模型。6周后观察动物生活状态、血液动力学和脏器/体重比的改变。结果模型组动物皮毛杂乱无光泽、气喘和活动减少;左室舒张压(LVDP)、左室舒张末压(LVEDP)均显著升高(P<0.01);+dp/dtmax和-dp/dtmax的绝对值均显著降低(P<0.01);心脏/体重、肺脏/体重、肝脏/体重比值显著增加(P<0.01)。结论豚鼠皮下注射异丙基肾上腺素6周,可有效建立豚鼠心肌缺血型CHF模型。 Objective To investigate the method of establishing animal model of medication-induced guineapig chronic heart failure. Methods The chronic heart failure models of guinea-pig were made by repetitious hypodermic isoproterenol, feeding in the same house with normal guinea pigs. Six weeks later, indexes including clinical features, hemodynamics, and the mass ratio of heart, lung, and liver to body were measured. Results The models showed untidy and lusterless fur, wheeziness and reduced activities. Left ventricular diastolic pressure (LVDP) and left ventricular end diastolic pressure (LVEDP) were significantly increased (P〈0.01). The absolute value of +dp/dt max and dp/dt max was significantly reduced (P〈0.01), The mass ratio of heart/body, lung/body and liver/body was significantly increased (P〈0.01). Conclusion The animal model of guinea-pig chronic heart failure may be formed by hypodermic injection of isoproterenol for 6 weeks.
