摘要
目的:研究全反式维甲酸(ATRA)对儿童卵黄囊瘤细胞诱导分化过程中细胞形态及维甲酸β受体表达的影响。方法:在体外细胞培养的基础上,观察细胞形态学的变化、细胞增殖动力学的变化,采用MTT比色法测定细胞细胞增殖活性,以半定量RT-PCR方法检测维甲酸β受体(RAR-β)mRNA的表达。结果:全反式维甲酸能使卵黄囊瘤细胞形态趋向良性分化,有效地抑制了肿瘤细胞增殖;半定量RT-PCR结果显示,经不同浓度的ATRA处理后的卵黄囊瘤细胞的RAR-βmRNA表达水平均上调。结论:ATRA对卵黄囊瘤细胞有诱导分化作用,ATRA通过上调维甲酸β受体基因表达实现其诱导分化作用。
Objective:to investigate the effect of all-trans-retinoic acid on proliferation of yolk sac tumor cell line and expression of RAR-β gene in Inducing differentiation in vitro, Methods:Yolk sac tumor cell line was exposed to all-trans retinoic acid. The morphological and functional changes of the cells were observed with MTT, the PAR-β mRNA expression levels were determined by semi-quantitative RTPCR. Results:After yolk sac tumor cells were treated with all-trans retinoic acid, the malignant phenotypes of the cells gradually changed to the muture cells.The proliferation index decreased markedly. The expression of the RAR-β mRNA increased dramatically, Conclusion:ATRA can inhibit proliferation and induce differentiation of yolk sac tumor cell line ,and PAR-β can be the induce-differentiation mechanism of ATRA on yolk sac tumor cell line in Vitro.
