摘要
目的研究环氧合酶-2抑制剂SC236对胃癌转移的影响,并初步探讨其作用机制。方法用人胃癌细胞株SGC7901建立裸鼠人胃癌原位移植瘤模型,随机分为实验组和对照组,分别经尾静脉注射SC236和生理盐水,观察各组生存期和肿瘤转移情况,并通过免疫组化方法检测移植瘤MVD、VEGF及MMP-2蛋白的表达。结果与对照组相比,SC236可明显延长荷瘤鼠生存期,抑制胃癌转移,降低移植瘤MVD,减低移植瘤VEGF及MMP-2蛋白的表达。结论SC236能有效抑制胃癌的转移,其机制可能与其抑制肿瘤的血管生成、减低MMP-2的表达有关。
Objective To study the effect of cox-2 inhibitor on tumor metastasis of an orthotopic implantation metastatic model. Methods BALB/C-nu/nu nude mice were implanted orthotopically using SGC7901 human gastric cancer intact tissue and were di vided into 2 groups at random.control and SC236. The survival period, the sizes and the numbers of the lesions were observed. Immunohistochemical methord was used to detect the MVD,VEGF and MMP-2 expression in implanted tumors. Results SC236 prolonged the survival period of tumor-loading nude mice compared with the control(P〈0.05 ). It could obviously inhibit the growth of the metastatic tumors ,and it could decrease the MVD, the expression of VEGF and MMP-2 in metastatic tumors. Conclusion SC236 has obviously antitumor activity on metastasis with human gastric cancer cell line in nude mice . The mechanism may be inhibit the angiogenesis and down-regulation of the expression of VEGF and MMP 2.
