摘要
目的:探讨CXCR4单克隆抗体对人乳腺癌细胞MCF-7凋亡的影响。方法:以CXCR4单克隆抗体作用于MCF-7细胞,通过DNA梯状电泳、Hochest33258/PI荧光染色进行凋亡定性分析,以Annexin V/PI双染法定量检测MCF-7细胞的凋亡。结果:与对照组相比,CXCR4单克隆抗体作用后MCF-7细胞出现凋亡,凋亡率明显升高。结论:CXCR4单克隆抗体明显促进MCF-7细胞的凋亡,提示CXCR4/SDF-1生物学轴对乳腺癌细胞MCF-7的凋亡也有重要影响。
Objective:To study the effect of CXCR4 monoclonal antibodies ( CXCR4 mAb) on apoptosis in human breast cancer cell line MCF -7. Methods: After the addition of CXCR4 mAb, the apoptosis in MCF -7 cell were measured with DNA ladder, Hochest 33258/PI, and Annexin V/PI analysis. Results: As compared with control group, the apoptosis occurred and the apoptotic rate significantly increased in MCF - 7 cell after treatment of CXCR4 mAb. Conclusion: The results suggested that CXCR4 mAb may induce apoptosis of MCF - 1 cell line and CXCRd / SDF - 1 may play an important role in regulating the apoptosis of MCF -7 cell.
