广西金秀瑶族D3S1358等6个STR基因座遗传多态性研究 被引量:1

摘要 目的了解广西金秀瑶族6个STR基因座(D3S1358、D5S818、D7S820、D13S317、FGA、vWA)的遗传多态性,探讨该民族与广西融水苗族等12个民族群体遗传关系。方法枸橼酸钠抗凝的广西金秀瑶族无相关健康个体血样175例,先用Chelex-100方法提取DNA,然后用AmpFeSTR Identifiler^TM kit荧光标记复合PCR扩增技术进行6个STR基因座复合扩增,用ABI3100型遗传分析仪对扩增产物进行电泳,并用GeneScan Analysis 3.7和Genetyper 3.7软件对扩增结果进行分析。利用D3S1358等6个STR基因座的基因频率计算广西金秀瑶族与广西融水苗族等12个民族群体间的遗传距离,并通过MEGA3.1软件的Neighbor—Joining方法构建系统发生树。结果广西金秀瑶族6个位点中检测出52种等位基因,基因频率分布在0.0029-0.3943之间;6个STR基因座的杂合度分布在0.7371—0.8686之间,个体识别力(DP)分布在0.8753—0.9601之间,累积个体识别力TDP为〉0.999999711,非父排除率(EP)在0.6046—0.8519之间,累积非父排除率CEP为0.999556633,多态信息量PIC分布在0.6788—0.8428之间。广西金秀瑶族等13个群体的系统发生树结果:广西金秀瑶族先跟广东瑶族和广西融水苗族相聚,然后与广西水族、广西汉族、广西京族、广西仫佬族、广西毛南族、广西临桂瑶族和广西三江侗族相聚,接着先后与广西崇左壮族、广东汉族相聚,最后与蒙古族相聚。结论广西金秀瑶族群体D3S1358等6个STR基因座均属于高度多态性遗传标记:广西金秀瑶族可能是以瑶族的遗传标记为主的多族源群体。 Objective To investigate the polymorphisms of 6 short tandem repeat(STR) loci(D3S1358,D5S818,D7S820,D13S317, FGA,vWA) in Yao ethitie group of Jinxiu in x Guangxi province,so as to reveal the genetic relationships among Yao ethnic group of Jinxiu as well as Miao eflmie group of Rongshtfi county and other twelve ethnic groups. Methods Sodium-eitrated blood specimens from 175 healthy genetically unrelated Yao individuals in Jinxiu were collected,The DNAs were extracted from the specimens by the methed of Chelex-100 extraction.6 STR loci in Yao ethnic group were screened by using the AmpFeSTR Identifiler ^TM PCR Amplification Kit and 3100 Genetic Analyzer,The data were compared with scanning the Genebank: Genescan Paralysis3.7 and Genetyper3.7. Polymorphisms and genetic distance were analysized base on Microsoft office excel statistial softwares. The phylogenetic analyses were made by Neighbor-Joining of MEGA3.1 software. Results 52 alleles of 6 STR loci were screened.The allele frequency ranged from 0.002 9 to 0.394 3.The heterezygosities(Ho) of 6 STR loci were between 0.737 1 and 0.868 6,with average heterezygosity being 0.786 7.The discrimination powers(DP) were from 0.875 3 to 0.960 1 and the exclusion probabilities of patenfity exclusion(EP) were from 0.604 6 to 0.851 9, restrictively, The cumulative exehtsion power(CEP) exceeded 0.999 556 633, together discrimination power(TDP) also exceeded 0.999 999 711, the polymorphism information contents ( PIC ) were from 0. 6788 to 0. 842 8. The analyses of phylogenetic tree showed that firstly Yao etlmie group of Jinxiu congregated with Miao ethnic group of Rongshui and Yao ethnic group of Guangdong; secondly congregated with other groups of Guangxi and Han ethnic group of Guangdong, lastly with Menggu ethnic group. Conclusions It's demonstrated that the polymorphisms of 6 STR loci exist in Yao ethnic group of Jinxiu in Guangxi province, which may be due to the fact the genetypes of Yao ethnic group in Jinxiu dominate those of any other ethnic groups in the same region.
