摘要
为测定人红细胞NADH-细胞色素b5还原酶(b5R)含量,建立了以兔抗b5R抗体为包被抗体,b5R单抗为显示抗体的双抗体夹心ELISA。利用此法,首次测得中国正常成人(n=30)红细胞胞浆的b5R含量为25.63±8.54ng/mgHb。新生儿、遗传性高铁血红蛋白血症患者及其家族半合子成员的红细胞胞浆b5R含量均低于正常成人水平,甚至低至测不出。结果证实,b5R酶蛋白量减少是新生儿和遗传性高铁血红蛋白血症患者红细胞b5R活性降低的机制之一。
